Tangier disease is characterized by very low blood plasma levels of HDL cholesterol, accumulation of cholesteryl esters in tissues, and an increased risk for developing cardiovascular disease. Unlike Barth syndrome, Tangier disease is mainly caused by abnormal enhanced production of CL. Studies show that there are three to fivefold increase of CL level in Tangier disease. Because increased CL levels would enhance cholesterol oxidation, and then the formation of oxysterols would consequently increase cholesterol efflux. This process could function as an escape mechanism to remove excess cholesterol from the cell.

== Medical uses == As a treatment (oral or injection), it is used to treat or prevent pyridoxine deficiency, sideroblastic anaemia, pyridoxine-dependent epilepsy, certain metabolic disorders, side effects of isoniazid treatment and certain types of mushroom poisoning. Isoniazid is an antibiotic used for the treatment of tuberculosis. Its common side effects include numbness in the hands and feet. Co-treatment with vitamin B6 alleviates the numbness. Pyridoxine-dependent epilepsy is a type of rare infant epilepsy that does not improve with typical anti-seizure medications. Pyridoxine in combination with doxylamine is used as a treatment for morning sickness in pregnant women.
=== Hormone excess === If there is an excess of growth hormone, it is usually because of over-secretion of somatotrope cells in the anterior pituitary gland. A significant amount of excess somatotrope secretion before puberty or before the end of new bone tissue growth can lead to gigantism, a disease that causes excess growth of body (e.g. being over 7 ft. tall) and unusually long limbs. An excess of secretion of growth hormone after puberty can lead to acromegaly. This is a disease that causes abnormal growth in the hands, head, jaw, and tongue. Some symptoms associated with acromegaly include heavy sweating, oily skin, improper processing of sugars in the diet (diabetes), high blood pressure, increased calcium in urine and swelling of the thyroid gland and arthritis.
