Everything below concerns phase 2 trial. We keep the language plain, cite what the science says, and separate well-supported claims from open questions.

Interpretation depends on study phase and duration. Phase 2 programs are powered for weight and safety signals, not for cardiovascular or renal outcomes, which require event-driven designs. Gastrointestinal events such as nausea, diarrhea, vomiting, and constipation are the most frequently reported adverse effects and tend to cluster around dose escalation. Small increases in heart rate have been described. Because follow-up after treatment discontinuation is limited, questions about weight regain and durability are open rather than answered.
Trial reports for this compound rely on a small set of repeated measures. Body weight is normally expressed as percent change from baseline at a fixed week, with absolute kilograms given secondarily. Glycemic endpoints include HbA1c, fasting glucose, and, in some protocols, continuous glucose monitoring summaries. Imaging endpoints such as MRI-derived proton density fat fraction quantify liver fat. Standardization matters because a percent change and a categorical responder analysis can tell different stories about the same dataset.
Body composition is assessed with dual-energy X-ray absorptiometry or comparable methods, which separate fat mass from lean mass. Reported losses include both compartments, and the ratio between them is a subject of ongoing analysis rather than a settled result. Waist circumference, blood pressure, and lipid panels are collected as supporting measures. Resting energy expenditure and substrate oxidation are measured in smaller mechanistic studies, where glucagon receptor activity is expected to matter. These substudies are typically short and small, so their findings carry wide uncertainty.
Development has progressed from single- and multiple-ascending-dose studies in healthy volunteers into larger randomized trials in adults with obesity, type 2 diabetes, and fatty liver disease. Early reports describe dose-dependent reductions in body weight and improvements in glycemic markers over treatment periods of several months. Whether the glucagon arm adds tolerability cost without added benefit is still debated. Long-term cardiovascular outcomes, effects after treatment stops, and performance in older or comorbid populations are open questions rather than settled findings. Approval status may change, so the current investigational label should be confirmed against regulatory sources.
Retatrutide is an investigational synthetic peptide developed under the code LY3437943, with a backbone derived from glucose-dependent insulinotropic polypeptide. Several non-proteinogenic residues, including alpha-aminoisobutyric acid, appear in that backbone, and a fatty diacid side chain attached through a linker extends circulation time. The molecule carries roughly thirty-nine amino acid units and a total mass near 4.7 kilodaltons. Administration is by subcutaneous injection once weekly. Published work uses both the name retatrutide and the code LY3437943.
Pharmacologically the compound activates three receptors: GLP-1, GIP, and glucagon. GLP-1 and GIP signaling contribute to glucose-dependent insulin release, delayed gastric emptying, and reduced appetite, while glucagon receptor activation is associated with increased energy expenditure and hepatic fat oxidation. The single-molecule design is intended to keep these activities in one peptide rather than combining separate agents. Relative activity at each receptor differs, and the balance between them is a central question in interpretation. The glucagon component is partly offset by incretin-mediated insulin secretion, an interaction that remains incompletely characterized.
| Property | Value | Notes |
|---|---|---|
| Trial design | Randomized, double-blind, placebo-controlled | Frequently includes dose-escalation arms |
| Weight endpoint | Percent change from baseline | Reported at a prespecified week |
| Liver fat endpoint | MRI proton density fat fraction | Non-invasive imaging measure |
| Body composition method | Dual-energy X-ray absorptiometry | Separates fat mass from lean mass |
| Common adverse events | Nausea, diarrhea, vomiting | Reported most often during dose escalation |
Several questions remain unresolved. It is not yet known whether the compound reduces cardiovascular events or mortality, because outcome studies require long follow-up. The durability of weight reduction after treatment withdrawal is uncertain, and rebound has been observed with other incretin-based therapies. Long-term safety data covering several years are limited. Effects in adolescents, in pregnancy, and in people with significant kidney or liver impairment have not been characterized in published reports.
Randomized studies of retatrutide measure change in body weight as a percentage of baseline, along with absolute weight loss. Glycemic endpoints include hemoglobin A1c and fasting plasma glucose. Investigators also track blood pressure, lipid fractions, and liver fat content to characterize effects beyond weight alone. Trial designs typically use double-blind, placebo-controlled groups with periodic dose escalation, and they record adverse events throughout both treatment and follow-up periods.
Quantification of the peptide in biological samples generally relies on liquid chromatography coupled with tandem mass spectrometry. This approach separates the analyte from matrix components and detects it by mass-to-charge transitions specific to the molecule. Immunoassays offer higher throughput but can cross-react with related peptides and metabolites, so mass spectrometric methods are preferred when structural confirmation is required. Method validation typically addresses accuracy, precision, selectivity, and stability under handling conditions.
The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.
Identity and purity are established by instrumental methods rather than by appearance. Reversed-phase high-performance liquid chromatography separates the peptide from related impurities and yields a purity value, usually expressed as the share of total peak area. Mass spectrometry checks that the observed mass agrees with the mass calculated from the published sequence, while peptide mapping or amino acid analysis adds structural evidence. Water content, counter-ion identity and residual solvents are sometimes reported as well. A certificate of analysis should name the method behind each figure, because results are method-dependent.
Laboratory handling follows the conventions used for other synthetic peptides. Lyophilized material is weighed and dissolved in an aqueous diluent, typically sterile water or bacteriostatic water, using gentle swirling rather than vigorous shaking, because foaming stresses the chain. Solutions are prepared under clean conditions and, where sterility matters, passed through a suitable filter. Working portions are kept small so that stock material is not repeatedly warmed and cooled, a practice that limits both aggregation and gradual loss of activity.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
=== Agents of spontaneous missense mutation === Environmental mutagens, such as tobacco smoke or UV radiation, may be a cause of spontaneous missense mutations. Tobacco smoke has been implicated in transversion mutations in the K-ras gene, with a meta-analysis of lung carcinomas showing 25 tumours containing a G to T mutation causing an amino acid change from glycine to cysteine, and 11 tumours with a G to T mutation causing an amino acid change from glycine to valine. Similarly, numerous studies have shown ultraviolet light induces missense mutations in the p53 gene, which when unregulated, reduces the cell's ability to recognize DNA damage and engage in apoptosis, leading to cell proliferation and potential skin carcinogenesis.
=== Human physiology === Potassium is the eighth or ninth most common element by mass (0.2%) in the human body, so that a 60 kg adult contains a total of about 120 g of potassium. The body has about as much potassium as sulfur and chlorine, and only calcium and phosphorus are more abundant (with the exception of the ubiquitous CHON elements). Potassium ions are present in a wide variety of proteins and enzymes. 98% of the potassium in a human body is inside of individual cells.
Setiptiline acts as a norepinephrine reuptake inhibitor, α2-adrenergic receptor antagonist, and serotonin receptor antagonist, likely at the 5-HT2 subtypes, as well as an H1 receptor inverse agonist/antihistamine.
Cordycepin, or 3'-deoxyadenosine, is a derivative of the nucleoside adenosine, differing from the latter by the replacement of the hydroxy group in the 3' position with a hydrogen. It was initially extracted from the fungus Cordyceps militaris, but can now be produced synthetically.
== Evolution == Based on parsimony-based phylogenetic analysis, the most recent common ancestor of all eukaryotes most likely already possessed an early RNAi pathway; the absence of the pathway in certain eukaryotes is thought to be a derived characteristic. This ancestral RNAi system probably contained at least one Dicer-like protein, one Argonaute, one PIWI protein, and an RNA-dependent RNA polymerase that may also have played other cellular roles. A large-scale comparative genomics study likewise indicates that the eukaryotic crown group already possessed these components, which may then have had closer functional associations with generalized RNA degradation systems such as the exosome. This study also suggests that the RNA-binding Argonaute protein family, which is shared among eukaryotes, most archaea, and at least some bacteria (such as Aquifex aeolicus), is homologous to and originally evolved from components of the translation initiation system.

=== Membership policy === WOSM's membership consists of its remaining founding member organizations and organizations recognized by WOSM as national scout organizations. WOSM's rules protect its founding and existing member organizations by permitting only one member organization in each country and locking-out all other Scout organizations from WOSM membership, recognition and participation no matter how worthy or large their membership. Several member organizations are federations, some with different component groups divided on the basis of religion (e.g., France and Denmark), ethnic identification (e.g., Israel) or language (e.g., Belgium). However, WOSM has never required an existing member organization to federate with other Scout organizations in the country, in order to make WOSM more inclusive and representative. There are numerous Canadian Scout organizations but only one is a WOSM member organization (the Canadian branch of one of the organizations that founded WOSM) which has a French language affiliate which is thereby recognized by WOSM. Other than this inherent limitation on WOSM membership, the basis for WOSM membership includes adherence to WOSM's aims and principles and independence from political involvement on the part of each member organization.
In 1912, Frederick Gowland Hopkins demonstrated that unknown accessory factors found in milk, other than carbohydrates, proteins, and fats were necessary for growth in rats. Hopkins received a Nobel Prize for this discovery in 1929. By 1913, one of these substances was independently discovered by Elmer McCollum and Marguerite Davis at the University of Wisconsin–Madison, and Lafayette Mendel and Thomas Burr Osborne at Yale University. McCollum and Davis ultimately received credit because they submitted their paper three weeks before Mendel and Osborne. Both papers appeared in the same issue of the Journal of Biological Chemistry in 1913. The "accessory factors" were termed "fat soluble" in 1918, and later "vitamin A" in 1920. In 1919, Harry Steenbock (University of Wisconsin–Madison) proposed a relationship between yellow plant pigments (β-carotene) and vitamin A. In 1931, Swiss chemist Paul Karrer described the chemical structure of vitamin A. Retinoic acid and retinol were first synthesized in 1946 and 1947 by two Dutch chemists, David Adriaan van Dorp and Jozef Ferdinand Arens.
Unlike classes I and II, Class III molecules have physiological roles and are encoded between classes I and II on the short arm of human chromosome 6. Class III molecules include several secreted proteins with immune functions: components of the complement system (such as C2, C4, and B factor), cytokines (such as TNF-α, LTA, and LTB), and heat shock proteins.
According to biographer Ian Stewart, by the mid-1990s Anwar's leadership ambition was the most serious threat to Mahathir's power. Anwar began to distance himself from Mahathir, overtly promoting his superior religious credentials and suggesting loosening the restrictions on civil liberties. However, Mahathir continued to back Anwar as his successor until the collapse of their relationship during the Asian financial crisis, with Mahathir abandoning the tight monetary and fiscal policies urged by the IMF. As Mahathir took the reins of Malaysia's economic policy over 1998, Anwar was increasingly sidelined. On 2 September, he was dismissed as deputy prime minister and finance minister and promptly expelled from UMNO. No immediate reasons were given for the dismissal. However, the media speculated that it related to lurid allegations of sexual misconduct circulated in a "poison pen letter" at the general assembly. As more allegations surfaced, large public rallies were held in support of Anwar. On 20 September, he was arrested and placed in detention under the Internal Security Act. Anwar stood trial on four charges of corruption, arising from allegations that Anwar abused his power by ordering police to intimidate persons who had alleged Anwar had sodomised them. He was found guilty in April 1999 and sentenced to six years in prison. In another trial shortly after, Anwar was sentenced to another nine years in prison on a conviction for sodomy. The sodomy conviction was overturned on appeal after Mahathir left office.

Tulips (Tulipa) also have a long cultural tradition, particularly in the Islamic world. The Persian word for tulips, lâleh (لاله), was borrowed into Turkish and Arabic as lale. In Arabic letters, lale is written with the same letters as Allah, and is used to denote God symbolically. Tulips became widely used in decorative motifs on tiles, fabrics, and ceramics in Islamic art and the Ottoman Empire in particular, and were revered in poetry, such as that of Omar Khayam in the twelfth century. Tulips were an essential part of the gardens of Persia, having been cultivated from the Steppes. As the Seljuks – Oghuz-Turkic leaders originated from the steppes – moved to Persia, and then west, they took tulips with them to Turkey, where many escaped cultivation and became naturalised. Today there are a number of places in Turkey called Laleli (‘with tulips’). Tulips first appeared in the decorative arts in Turkey in the thirteenth century and flourished under the Ottomans, in particular in the royal palaces, and was adopted by the Osmans as their symbol. Further species were collected from Persia and the spreading Ottoman Empire saw to it that tulip culture also spread. By the sixteenth century it was a national symbol, hence the designation "Tulip era", by which time they were becoming of economic importance.
=== Diet === For most horses, diet has a significant impact on the degree of clinical signs. PSSM horses fed diets high in nonstructural carbohydrates (NSC), which stimulate insulin secretion, have been shown to have increased severity of rhabdomyolysis with exercise. Current recommendations for horses with PSSM include a low-starch, high-fat diet. Low-starch diets produce low blood glucose and insulin levels after eating, which may reduce the amount of glucose taken up by the muscle cells. High fat diets increase free fatty acid concentrations in the blood, which may promote the use of fat for energy (via free fatty acid oxidation) over glucose metabolism. Horses with the most severe clinical signs often show the greatest improvement on the diet. Dietary recommendations usually include a combination of calorie restriction, reduction of daily NSC content, and an increase in dietary fat. Diet recommendations need to be balanced with the animal's body condition score and exercise level, as it may be beneficial to wait on increasing dietary fat after an obese animal has lost weight. The diet should have <10% of digestible energy coming from NSC, and 15–20% of daily digestible energy coming from fat.
Fourth, and more debated, article 24 requires that the longer an EU citizen stays in a host state, the more rights they have to access public and welfare services, on the basis of equal treatment. This reflects general principles of equal treatment and citizenship in TFEU articles 18 and 20. In a simple case, in Sala v Freistaat Bayern the Court of Justice held that a Spanish lady who had lived in Germany for 25 years and had a baby was entitled to child support, without the need for a residence permit, because Germans did not need one. In Trojani v Centre public d'aide sociale de Bruxelles, a French man who lived in Belgium for two years was entitled to the "minimex" allowance from the state for a minimum living wage. In Grzelczyk v Centre Public d'Aide Sociale d'Ottignes-Louvain-la-Neuve a French student, who had lived in Belgium for three years, was entitled to receive the "minimex" income support for his fourth year of study. Similarly, in R (Bidar) v London Borough of Ealing the Court of Justice held that it was lawful to require a French UCL economics student lived in the UK for three years before receiving a student loan, but not that he had to have additional "settled status". Similarly, in Commission v Austria, Austria was not entitled to restrict its university places to Austrian students to avoid "structural, staffing and financial problems" if (mainly German) foreign students applied, unless it proved there was an actual problem.
==== Desenrola Brasil ==== In July 2023, the Ministry of Finance launched the first phase of the "Emergency Debt Renegotiation Programme for Indebted Individuals" (popularly known as the Desenrola Brasil programme), a debt renegotiation programme which was divided into two phases. In October of the same year, the second phase, in which non-bank debts were renegotiated, was launched. Desenrola Brasil is made up of four "participants" (the federal government, debtors, creditors (such as financial institutions, public utility services, retail companies, service providers in general, including individual microentrepreneurs and small businesses), and financial agents, such as banks) and has the stated objective of reducing debt among the population (especially the low income and middle classes), as well as facilitating access to credit by the general population.

A responder analysis counts participants who cross a threshold, such as five or ten percent weight loss. It complements average percent change by showing how widely results are distributed. The two measures can diverge when a subset of participants loses a large amount.
Mechanistic measures such as energy expenditure and liver fat change come from small substudies and should be read as preliminary. Long-term clinical outcomes are not yet available. Average weight loss figures are better supported than explanations for how the loss is achieved.
Endpoints reported at different weeks, in different baseline BMI ranges, or under different dose-escalation schedules are not directly comparable. Studies in type 2 diabetes often show smaller weight changes than studies in obesity without diabetes. Reporting the population alongside the number keeps comparisons honest.
As of the mid-2020s retatrutide remains investigational and is not an approved medicine in the United States or the European Union. It has been supplied mainly to participants in clinical trials. Labels and availability can change, so regulatory listings should be checked directly.
