Identification and purity assessment rely on established analytical techniques. Reverse-phase high-performance liquid chromatography separates the compound from related impurities and degradation products. Mass spectrometry confirms molecular identity and detects modifications that change the expected mass. Additional methods such as amino acid analysis or capillary electrophoresis may be used for verification. Small differences in sample preparation can influence results, so procedures are usually controlled and documented in detail. Consistency between runs supports confidence in reported values.
Stability studies examine how the molecule changes under defined conditions of temperature, humidity, and light exposure over time. Results are used to set storage recommendations and shelf-life limits. In practice, lyophilized peptide material is often stored at low temperatures to slow degradation, while reconstituted solutions are handled more carefully because they are generally less stable. Reported stability data apply to specific formulations and conditions, so extrapolation to other preparations requires caution.

Peptide content and purity are commonly measured by reversed-phase high-performance liquid chromatography with ultraviolet detection, using gradient elution over a C18 column. Identity is confirmed by mass spectrometry, because the theoretical monoisotopic mass allows unambiguous assignment of the main component. Impurity profiling resolves deletion sequences, oxidized residues, and truncated fragments. Since the molecule carries a lipophilic side chain, mobile phases often include ion-pairing agents and organic modifiers to keep peaks symmetric.
Lyophilized material is generally held at minus 20 degrees Celsius or colder for long-term storage, protected from moisture and light. Solutions are handled under refrigeration, typically between 2 and 8 degrees Celsius, and used within a short window because degradation and microbial growth both accelerate in liquid. Repeated freeze-thaw cycles are avoided, and vials are equilibrated to room temperature before opening to reduce condensation. These are general laboratory conventions for peptides of this size rather than product-specific directions.
Characterization panels may add amino acid analysis for compositional confirmation, circular dichroism for secondary structure in solution, and light scattering for aggregation tendency. Aggregation is a central concern for peptides bearing hydrophobic side chains, since it can lower measured potency and complicate accurate dosing. Stability studies examine temperature, humidity, pH, and light exposure over defined intervals, reporting the percentage of intact peptide remaining. Results depend strongly on the assay used, so comparing values across studies requires matching method details.
Solid material is generally held at -20 °C or colder, while reconstituted solutions are kept at 2-8 °C and used within a short window. Buffers that maintain a slightly acidic to neutral pH tend to improve short-term peptide stability. Repeated warming and cooling of stock solutions promotes aggregation and should be avoided. Container closures should remain intact, since adsorption to some plastics can reduce the amount of peptide in solution.
Identity and purity are established with reversed-phase high-performance liquid chromatography and mass spectrometry. Chromatographic profiles reveal related impurities, truncated sequences, and oxidation products, while mass measurement confirms the expected molecular mass. Purity values for research material are typically reported as a percentage by peak area. Reference standards help calibrate retention behavior across instruments. Independent laboratories emphasize method suitability because results depend heavily on column chemistry, gradient, and detection wavelength. Batch-to-batch comparison relies on the same validated method.
Investigational peptide material is commonly distributed as a lyophilized powder in sealed vials. The solid form appears as a white to off-white cake or powder and is hygroscopic once opened. Peptides of this size are sensitive to moisture, repeated freeze-thaw cycles, and prolonged exposure to ambient light. Handling practices therefore emphasize desiccation, minimal vial opening, and cold storage. Working aliquots are often prepared to avoid repeatedly warming the bulk container.
A presentation seen during gameplay reveals that GLaDOS was included in a proposed bid for de-icing fuel lines, incorporated as a fully functional disk-operation system that is arguably alive, unlike Black Mesa's proposal, which inhibits ice, nothing more. After contracting a terminal illness from moon rocks used in portal experiments, Cave decides to attempt to back up his consciousness to a computer. He realises that the technology will not be ready in time to backup his brain due to his imminent demise, and leaves instructions to forcefully backup the consciousness of his assistant Caroline. Upon being uploaded to GLaDOS, Caroline attempts to kill every scientist in the building in under a fraction of a second. After extensive attempts to control her with smaller robots called personality spheres that would act as emotional limits and a sort of artificial "conscience", GLaDOS managed to eventually convince the scientists that she was primarily concerned with science and requested a lethal brain neurotoxin for an experiment about cats. GLaDOS initiated the experiment during the company's first annual bring-your-daughter-to-work day in 1998. Immediately after activation, the facility was flooded with deadly neurotoxin by the AI. Events of the first Half-Life game occur shortly after that, presumably leaving the facility forgotten by the outside world due to apocalyptic happenings. Wolpaw, in describing the ending of Portal 2, affirmed that the Combine invasion, chronologically taking place after Half-Life and before Half-Life 2, had occurred before Portal 2's events.
Ethylene oxide is one of the most commonly used sterilization methods in the healthcare industry because of its non-damaging effects on delicate instruments and devices that require sterilization, and for its wide range of material compatibility. It is used for instruments that cannot tolerate heat, moisture, or abrasive chemicals, such as electronics, optical equipment, paper, rubber, and plastics. It is the most common chemical sterilization method, used for approximately 70% of total sterilizations, and for over 50% of all disposable medical devices. Ethylene oxide treatment was developed in the 1940s as a sterilant by the US military, and its use as a medical sterilant dates to the late 1950s, when the McDonald process was patented for medical devices. The Anprolene system was patented in the 1960s by Andersen Products, and it remains the most commonly used system in several niche markets, notably the veterinary market and some international markets. It relies on the use of a flexible sterilization chamber and an EtO cartridge for small volume sterilization, and where environmental and/or portability considerations dictate the use of a low dose. It is therefore referred to as the "flexible chamber sterilization" method, or the "gas diffusion sterilization" method. In the United States, the operation of EtO sterilization is overseen by the EPA through the National Emissions Standards for Hazardous Air Pollutants (NESHAP).
Hans Keller (11 March 1919 – 6 November 1985) was a Viennese Jewish British musician and prolific writer, who made significant contributions to musicology and music criticism; best known for his appearance on TV show The Look of the Week in which he interviewed Syd Barrett and Roger Waters. Keller was generally puzzled by, or even contemptuous of, the group and its music, opening with the comment "why has it all got to be so terribly loud?" Judith Kerr, OBE (surname pronounced /ˈkɑːr/ KAR German pronunciation: [kɛʁ]; 14 June 1923 – 22 May 2019) was a German-born British writer and illustrator whose books sold more than 10 million copies around the world. She created both enduring picture books such as the Mog series and The Tiger Who Came to Tea and novels for older children such as the semi-autobiographical When Hitler Stole Pink Rabbit, which gave a child's-eye view of escaping Hitler's persecution in the Second World War. Born in the Weimar Republic, she came to Britain with her family in 1935 to escape persecution during the rise of the Nazis. Gerald Kersh, novelist Beeban Kidron, Baroness Kidron, (b.
=== Prenatal development === The prenatal portion of tissue-resident dermal macrophages is produced from yolk-sac derived precursors. The generation of dermal macrophages results from primitive haematopoiesis or definitive haematopoiesis. Primitive haematopoiesis allows the generation of yolk-sac derived macrophages and subsequent release into the foetal bloodstream for tissue infiltration and colonisation. The infiltration of the skin by yolk-sac derived macrophages occurs as soon as 8.5 days after fertilisation. Different gene expressions regulate this process. It is independent of the MYB gene and dependent on the PU.1 gene. Definitive haematopoiesis occurs from 11 days and onwards after fertilisation. Monocytes are produced from haematopoietic stem cells in the liver, and they subsequently infiltrate the skin via the foetal bloodstream. In this process, cytokines like CSF1 are essential in facilitating the differentiation of monocytes into tissue-resident dermal macrophages and their survival. Therefore, the differentiation of monocytes to dermal macrophages is CSF-1/CSF1R dependent. The level of dermal macrophages from prenatal development remains detectible through constant, slow proliferation.

This hypothesis was opposed by French palaeontologist Marcellin Boule, who authored several publications starting in 1908 describing the French Neanderthal specimen La Chapelle-aux-Saints 1 ("The Old Man") as a slouching, ape-like creature distantly related to modern man. Boule's ideas would define discussions of Neanderthals for some time. Boule suggested two different lineages existed in Ice Age Europe: a more evolved one descending from the British Piltdown Man (a hoax) to the French Grimaldi Man (a Cro-Magnon) which would culminate with modern Europeans; and a less evolved dead-end lineage leading from the German Heidelberg Man to Neanderthal Man. As the focus of human origins shifted from Europe to East Asia ("Out of Asia" hypothesis) by the 1930s and 40s with discoveries such as Java Man and Peking Man (as well as the marginalisation of Piltdown Man), the question of a "Neanderthal phase" in human evolution once again became a topic of discussion. The definition of "Neanderthal" expanded to include several anatomically variable specimens around the Old World. In the contextual debate between monogenism and polycentrism, some specimens were described as "progressive" Neanderthals which would evolve into some local subspecies of H. sapiens (polycentricism), while the "classic" Neanderthals of the Western European Würm glaciation would not. In the 1970s, with the formulation of cladistics and the consequent refinement of the anatomical definitions of species, this "global morphological pattern" fell apart.

From the perspective of its commercial promoters, scientific breakthroughs, industrial commitment, and official support were finally coming together, and biotechnology became a normal part of business. No longer were the proponents for the economic and technological significance of biotechnology the iconoclasts. Their message had finally become accepted and incorporated into the policies of governments and industry.

A neurotransmitter sodium symporter (NSS) (TC# 2.A.22) is a type of neurotransmitter transporter that catalyzes the uptake of a variety of neurotransmitters, amino acids, osmolytes and related nitrogenous substances by a solute:Na+ symport mechanism. The NSS family is a member of the APC superfamily. Its constituents have been found in bacteria, archaea and eukaryotes.
As of 2018, the overwhelming majority of bitcoin transactions took place on cryptocurrency exchanges. Since 2014, regulated bitcoin funds also allow exposure to the asset or to futures as an investment. Bitcoin is used as a store of value: individuals and companies such as the Winklevoss twins and Elon Musk's companies SpaceX and Tesla have each bought and sold hundreds of millions of dollars worth of bitcoin. Bitcoin wealth is highly concentrated, with 0.01% holding 27% of in-circulation currency, as of 2021. As of March 2025, El Salvador had $550 million worth of bitcoin in its international reserves, about 6,102 coins. Bitcoin, along with other cryptocurrencies, has been described as an economic bubble by several economists, including Nobel Prize in Economics laureates, such as Joseph Stiglitz, James Heckman, and Paul Krugman. Another recipient of the prize, Robert Shiller, argues that bitcoin is rather a fad that may become an asset class. He describes its price growth as an "epidemic", driven by contagious narratives. In 2024, Jean Tirole, also Nobel laureate, described bitcoin as a "pure bubble" as its intrinsic value is zero. According to him, some bubbles are long-lasting such as gold and fiat currencies, and it is impossible to predict whether bitcoin would implode like other financial bubbles or become an alternative to gold. The same year, Federal Reserve Chair Jerome Powell described bitcoin as a digital competitor to gold but not to the dollar as he argued it is a highly volatile speculative asset not used as a form of payment.
An ideal beam-splitter transmits and reflects 50% of the incident radiation. However, as any material has a limited range of optical transmittance, several beam-splitters may be used interchangeably to cover a wide spectral range. In a simple Michelson interferometer, one beam passes twice through the beamsplitter but the other passes through only once. To correct for this, an additional compensator plate of equal thickness is incorporated. For the mid-IR region, the beamsplitter is usually made of KBr with a germanium-based coating that makes it semi-reflective. KBr absorbs strongly at wavelengths beyond 25 μm (400 cm−1), so CsI or KRS-5 are sometimes used to extend the range to about 50 μm (200 cm−1). ZnSe is an alternative where moisture vapour can be a problem, but is limited to about 20 μm (500 cm−1). CaF2 is the usual material for the near-IR, being both harder and less sensitive to moisture than KBr, but cannot be used beyond about 8 μm (1,200 cm−1). Far-IR beamsplitters are mostly based on polymer films, and cover a limited wavelength range.
AlphaFold 1 (2018) was built on work developed by various teams in the 2010s, work that looked at the large databases of related protein sequences now available from many different organisms (most without known 3D structures), to try to find changes at different residues (peptides) that appeared to be correlated, even though the residues were not consecutive in the main chain. Such correlations suggest that the residues may be close to each other physically, even though not close in the sequence, allowing a contact map to be estimated. Building on recent work prior to 2018, AlphaFold 1 extended this by estimating a probability distribution for the distances between residues, effectively transforming the contact map into a distance map. It also used more advanced learning methods than previously to develop the inference. The code was not made publicly available, except to run on sequences of proteins in the 2018 CASP competition.
