The peptide backbone is chemically modified to resist rapid enzymatic breakdown in the body. A fatty acid side chain promotes binding to serum albumin, which slows renal clearance and supports an extended circulation time. These modifications allow less frequent administration than would be possible with an unmodified peptide. The precise contribution of glucagon receptor activation to the overall metabolic effect remains an area of active investigation, because glucagon raises glucose while also increasing energy expenditure.
Development has progressed through early- and mid-stage human studies in adults with obesity and with type 2 diabetes. Published phase 2 data reported reductions in body weight and improvements in glycemic markers over the treatment period. No regulatory agency has approved the compound for any indication, and it remains available only within controlled research settings. Whether benefits observed in trials translate into durable outcomes after treatment stops is not yet established.
Retatrutide is an investigational synthetic peptide that acts as an agonist at three distinct G protein-coupled receptors. It combines activity at the glucagon-like peptide-1 receptor, the glucose-dependent insulinotropic polypeptide receptor, and the glucagon receptor within a single molecule. This multi-receptor profile distinguishes it from earlier incretin-based compounds that engage one or two of these pathways. Researchers designed the molecule to test whether simultaneous activation produces greater metabolic effects than single or dual agonism alone.

Solid peptide is generally held as a lyophilised powder at low temperature to slow degradation, with desiccant to limit moisture uptake. Reconstituted solutions are less stable and are usually kept refrigerated and protected from light for short periods. Repeated freeze-thaw cycles are avoided because they encourage aggregation. Laboratory handling includes work in a fume hood or laminar flow cabinet to limit inhalation and contamination. Weighing and transfer steps are performed with antistatic tools to reduce static-driven loss of fine powder.
Characterising a peptide of this size relies on a combination of chromatographic and mass spectrometric techniques. Reversed-phase high-performance liquid chromatography separates the intact molecule from related impurities, while electrospray mass spectrometry confirms molecular mass and detects truncation or oxidation products. Peptide mapping after enzymatic digestion verifies the amino acid sequence and locates modified residues. Because the molecule carries a lipid chain, assays must also distinguish the correctly conjugated product from incompletely acylated species.
Peptides in this class degrade mainly through hydrolysis, oxidation, and aggregation. The lipid modification improves plasma residence time but can also promote self-association in aqueous solution at higher concentrations. Oxidation of methionine and deamidation of asparagine residues are common chemical liabilities that accumulate during storage. Stability studies therefore track purity loss, aggregate formation, and changes in receptor-binding potency over time under defined temperature and humidity conditions.
Receptor activation produces downstream effects that differ by tissue. GLP-1 receptor signaling influences appetite regulation and insulin secretion in a glucose-dependent manner. GIP receptor activity contributes to metabolic handling of nutrients and may modulate adipose tissue. Glucagon receptor engagement raises energy expenditure and promotes hepatic lipid turnover, though the balance among these actions in humans is still being characterized. Preclinical models showed reductions in body weight and improved glycemic markers.
Clinical development has progressed through phase 2 trials in adults with obesity and type 2 diabetes, with phase 3 programs reported as ongoing. Reported outcomes include reductions in body weight and improvements in glycemic measures over defined treatment periods. Whether these effects translate into durable benefits after treatment ends remains an open question. Long-term safety data across broad populations are not yet complete, and regulatory decisions have not been announced.
Human evidence remains limited to controlled studies. A phase 2 trial in adults with obesity reported large, dose-dependent reductions in body weight over 48 weeks, with gastrointestinal events as the most frequently recorded adverse effect. Phase 3 programs designated TRIUMPH, for obesity, and TRANSCEND, for type 2 diabetes, are intended to confirm efficacy and to characterize safety in larger populations. Related studies are examining conditions such as knee osteoarthritis in people with obesity and metabolic liver disease. Open questions include long-term tolerability, effects on lean mass, and what happens after treatment is stopped.
Retatrutide is an investigational synthetic peptide that acts on three receptor targets at once: glucose-dependent insulinotropic polypeptide, glucagon-like peptide-1, and glucagon. It is developed by Eli Lilly and appears in the literature and in trial registries under the code LY3437943. The molecule belongs to a class of engineered peptides designed to resist rapid breakdown and permit infrequent subcutaneous administration. No regulatory agency has approved it for clinical use, and all available human data come from controlled trials rather than from routine practice.
Each receptor contributes a different physiological effect. Activation of the GLP-1 receptor slows gastric emptying and reduces appetite signaling in the brain. GIP receptor activity influences insulin secretion and lipid handling, while glucagon receptor stimulation raises energy use and fat oxidation. Combining these pathways is intended to produce weight loss beyond what single- or dual-receptor agonists achieve. Researchers attribute the observed potency to simultaneous engagement of all three targets, though the exact contribution of each receptor to overall effect remains under investigation.
Clinical development has advanced through phase 2 trials in adults with obesity and type 2 diabetes. Reported phase 2 results described substantial average weight reduction over roughly forty-eight weeks of weekly dosing. A phase 3 program is ongoing to confirm efficacy and assess long-term safety. Because the compound has not received regulatory approval, it is not available as a prescription product. Public discussion of retatrutide often conflates trial findings with marketed status, an important distinction when interpreting coverage of the topic.
== Endolithic parasitism == Until the 1990s phototrophic endoliths were thought of as somewhat benign, but evidence has since surfaced that phototrophic endoliths (primarily cyanobacteria) have infested 50 to 80% of midshore populations of the mussel species Perna perna located in South Africa. The infestation of phototrophic endoliths resulted in lethal and sub-lethal effects such as the decrease in strength of the mussel shells. Although the rate of thickening of the shells were faster in more infested areas it is not rapid enough to combat the degradation of the mussel shells.
Local botulinum toxin A injections for cosmetic purposes have on occasion caused weaknesses in distant muscles, symptoms resembling ocular or generalized myasthenia gravis in individuals with subclinical myasthenia gravis, and exacerbations of previously controlled myasthenia gravis. Botulinum toxin A has also been used to treat spasmodic torticollis (i.e., involuntarily neck turning), blepharospasm (involuntary contraction of the eye lids), and other uncontrolled facial muscle spasms in people with myasthenia gravis without side effects or with only short-lived dysphagia or diplopia. Magnesium: Magnesium is a chemical element that blocks skeletal muscle contraction by inhibiting the release or acetylcholine at the neuromuscular junction and also by lowering the sensitivity of these muscles to acetylcholine. Respiratory failure has occurred after systemic use of magnesium (mainly in the form of intravenous magnesium sulfate injections) for pre-eclampsia and after magnesium replacement during the course of a hospitalization in people with underlying myasthenia gravis. Local anesthetics: Local anesthetics cause absence of pain and all other sensations in a specific body part without loss of consciousness. There are two broad classes of these anesthetics: esters (i.e., procaine, cocaine, tetracaine benzocaine, and chloroprocaine) and amides (i.e. , lidocaine, bupivacaine, etidocaine, levobupivacaine, mepivacaine, prilocaine, and ropivacaine).
On 21 September, Pakistan launched airstrikes on the provinces of Paktika and Kunar in retaliation for the Kohat attack. Kunar's Nurgal district was hit at 4:00 local time, striking a family home and killing three family members and four others. Paktika's Barmal district was hit at 3:00, a shop and an unoccupied house were destroyed in the bombardment. Pakistani authorities denied striking civilians and said the attacks struck militant hideouts perfectly, supposedly killing 28 militants and multiple suicide bombers. On 22 September, it was reported that several Pakistani Taliban members attempted to cross the border at Kurram from Nangarhar and Paktia in Afghanistan. After being identified by Pakistani troops, they reportedly began firing on Pakistani checkposts. In retaliation, Pakistani troops launched multiple attacks on Afghan checkposts and defensive positions, supposedly causing several to flee. On 23 September, there were supposedly drone strikes inside Pakistan. While Pakistan blamed Afghanistan, Taliban officials denied the use of drones. On 24 September, Pakistan carried out airstrikes on 10 sites in Afghanistan. Kandahar, Paktia and Khost were struck and at least four civilians were killed. Afghanistan vowed to respond. The same day, local Afghan sources like Hariyat Media claimed that a Pakistani drone was intercepted over the Garmsir district near a Taliban border brigade position. On 27 September, Afghanistan's Defence Ministry claimed that 28 fighters were killed in Nuristan who had crossed the border from Pakistan.
Feminist Anne Koedt argued that because men "have orgasms essentially by friction with the vagina" and not the clitoral area, this is why women's biology had not been properly analyzed. "Today, with extensive knowledge of anatomy, with [C. Lombard Kelly], Kinsey, and Masters and Johnson, to mention just a few sources, there is no ignorance on the subject [of the female orgasm]", she stated in her 1970 article The Myth of the Vaginal Orgasm. She added, "There are, however, social reasons why this knowledge has not been popularized. We are living in a male society which has not sought change in women's role". Supporting an anatomical relationship between the clitoris and vagina is a study published in 2005, which investigated the size of the clitoris; Australian urologist Helen O'Connell, described as having initiated discourse among mainstream medical professionals to refocus on and redefine the clitoris, noted a direct relationship between the legs or roots of the clitoris and the erectile tissue of the bulbs and corpora, and the distal urethra and vagina while using magnetic resonance imaging (MRI) technology. While some studies, using ultrasound, have found physiological evidence of the G-spot in women who report having orgasms during vaginal intercourse, O'Connell argues that this interconnected relationship is the physiological explanation for the conjectured G-spot and experience of vaginal orgasms, taking into account the stimulation of the internal parts of the clitoris during vaginal penetration. "The vaginal wall is, in fact, the clitoris", she said.

The term in situ does not appear in Classical Latin. Its earliest recorded use is in Late Latin, with the first known instance in the writings of Augustine of Hippo (354–430 AD). It became widely used in Medieval Latin. In English, the earliest known usage dates to the mid-17th century; the Oxford English Dictionary cites the first appearance in 1648, in William Molins' anatomical text Myskotomia. The usages in scientific literature increased from the late 19th century onward, initially in medicine and engineering, including geological surveys and petroleum extraction. During this period, the term described analyses conducted within the living human body or inside oil wells, among other applications. In situ entered French medical discourse by 1877 in the Journal de médecine et de chirurgie pratiques ('Journal of Practical Medicine and Surgery'). The compound term carcinoma in situ, referring to abnormal cells that are confined to their original location without invasion of surrounding tissue, was first used in a 1932 paper by U.S. surgical pathologist Albert C. Broders. The concept of in situ in contemporary art developed in the late 1960s and 1970s as a framework for artworks created specifically for a given space. By the mid-1980s, the term was adopted in materials science, particularly in the field of heterogeneous catalysis, where a catalyst in one phase facilitates a chemical reaction in a different phase. Its usage later expanded beyond catalysis and is now applied across various disciplines within materials science.
=== Blood === A blood sample of approximately 10 ml (0.35 imp fl oz; 0.34 US fl oz) is usually sufficient to screen and confirm most common toxic substances. A blood sample provides the toxicologist with a profile of the substance that the subject was influenced by at the time of collection; for this reason, it is the sample of choice for measuring blood alcohol content in drunk driving cases.
Villages in the Jabal al-Druze have many historical and ancient churches, most of them dedicated to saints favored by the Arabs. The architecture of the Byzantine era was influenced by the spread of Christianity and the consequent construction of churches and monasteries, the majority dating between the 4th century and early 6th century.
A vault is a large ribonuclear protein particle, a membrane-less organelle, three times the size of a ribosome but with only three proteins in contrast to the near hundred in the ribosome. Most human cells have around 10,000 vaults, and in some types of immune cell there may be up to 100,000. Macrophages have the greatest number of vaults of any human cell. Vaults are largely overlooked because their functions are purely speculative. They may play a role in transport from the nucleus to the cytoplasm, and may serve as scaffolds for signal transduction proteins. They are present in normal tissues, and more so in secretory and excretory epithelial cells.
==== Down syndrome ==== Trisomy 21 or Down syndrome is the most common human chromosomal anomaly arising from abnormal chromosomal segregation in meiosis. The condition can occur during anaphase in meiosis(I) marking oocyte maturation before ovulation and/or during anaphase in meiosis (II) signifying fertilization. Metabolic impact during these stages is furthered by low vitamin B12. Methylation of homocysteine to methionine is affected, primarily by the (MTRR):c.66A>G polymorphism. Chronic homocysteine elevation increases s-adenosyl-L-homocysteine levels, consequently inhibiting methyltransferase activity and promoting DNA hypomethylation. Mothers homozygous for this mutation (GG phenotype) are at a greater risk of having a child with down syndrome compared to heterozygotes (GA phenotype). Geographically, Irish populations are more likely to be homogenous whilst north American populations are commonly heterogeneous, resulting in a greater incidence of the polymorphism in the former group. The homozygous mutant allele promotes DNA hypomethylation and meiotic non-disjunction, increasing the risk of down syndrome. This polymorphism correlates to a 2.5 fold risk increase independently and a 4 fold increase in risk when co-expressed with the 677C>T MTHFR mutation. Combination with the MTR2756A>G genetic polymorphism further elevates down syndrome risk.

Among the newly emergent strata were wealthy industrialists and financiers, distinct from the traditional, but still critically important landed aristocracy. The intelligentsia, an educated, professional or business middle class, often originated from lower gentry, landless or alienated from their rural possessions, and from urban people. Many smaller agricultural enterprises based on serfdom did not survive the land reforms. The industrial proletariat, a new underprivileged class, was composed mainly of poor peasants or townspeople forced by deteriorating conditions to migrate and search for work in urban centers in their countries of origin or abroad. Millions of residents of the former Commonwealth of various ethnic groups worked or settled in Europe and in North and South America. Social and economic changes were partial and gradual. The degree of industrialisation, relatively fast-paced in some areas, lagged behind the advanced regions of Western Europe. The three partitions developed different economies and were more economically integrated with their mother states than with each other. In the Prussian Partition, for example, agricultural production depended heavily on the German market, whereas the industrial sector of Congress Poland relied more on the Russian market.
=== International activities === Since 2021 when its restaurants in China closed, White Castle does not currently have any restaurants outside of the continental United States. Through franchise deals with local corporate business partners, White Castle briefly had restaurants outside of the United States in Singapore, Malaysia, and Japan during the late 1980s and early 1990s, but the concept never caught on in those countries. During the same time period, White Castle also tried to establish franchised operations in Mexico and South Korea, but these restaurants also failed. The lone Korean restaurant in Seoul was quietly closed by 1993. In 1986, White Castle opened its first Japanese restaurant in the city of Osaka via a franchise deal with a Japanese company. There are no reliable records that show when this location closed. By the end of 1986, the Japanese franchise had six restaurants with a seventh opened by the following year. In June 1989, White Castle and its local franchise partner Innovest Bhd. opened seven restaurants in Malaysia. Innovest franchise territory included Malaysia and Singapore, and the company had plans to open three more restaurants by the end of the year, with the possibility of having a total of 20 restaurants within its two country region by the following year. The first White Castle franchised location in Mexico opened in Mexico City in 1996, but it also closed after a brief trial run. In 2017, White Castle opened its first and second restaurants in China in the city of Shanghai through a partnership with Shanghai-based ClearVue Partners.
The first Lower Saxon parliament or Landtag met on 9 December 1946. It was not elected; rather it was established by the British Occupation Administration (a so-called "appointed parliament"). That same day the parliament elected the Social Democrat, Hinrich Wilhelm Kopf, the former Hanoverian president (Regierungspräsident) as their first minister-president. Kopf led a five-party coalition, whose basic task was to rebuild a state afflicted by the war's rigours. Kopf's cabinet had to organise an improvement of food supplies and the reconstruction of the cities and towns destroyed by Allied air raids during the war years. Hinrich Wilhelm Kopf remained – interrupted by the time in office of Heinrich Hellwege (1955–1959) – as the head of government in Lower Saxony until 1961. The greatest problem facing the first state government in the immediate post-war years was the challenge of integrating hundreds of thousands of refugees from Germany's former territories in the east (such as Silesia and East Prussia), which had been annexed by Poland and the Soviet Union. Lower Saxony was at the western end of the direct escape route from East Prussia and had the longest border with the Soviet Zone. On 3 October 1950 Lower Saxony took over the sponsorship of the very large number of refugees from Silesia. In 1950 there was still a shortage of 730,000 homes according to official figures.
